If I set up a new tank but use my current mature media filter, will this increase the cycle process? Or will it kill off my healthy bacteria?

no..if you use the water from your old tank aswell with a bit topped up this will fully cycle the tank. as long as the filter isnt without oxygen for more than 20 mins

Yup, keep the filter running in a tub or something like that so the filter stays alive & you're ready to run.
